How combinatorial indexing enables scalable single cell sequencing
- Patented split pool barcoding technology and combinatorial index library preparation methods leverage the cells themselves as containers.
- Isolated cells or nuclei are distributed into specialized plates, where an initial barcode is added to the cell or each molecule within the cell (such as cDNA , gDNA , RNA , and proteins)
- Barcoded cells / molecules are collected from the first plate, pooled and again randomly distributed to the next dedicated plate, where a second barcode is added. Cells to be profiled can be scaled up exponentially by repeated pooling and partitioning, enabling single-cell analysis of hundreds of thousands to millions of cells.
